Note: Some unlockables will not appear directly after you complete their requirements. A lot of times you will need to back out of your Driver License screen and reload it or simply restart the game. You should also know that most unlockables have an alternate way of being unlocked. These other methods generally meet by completing a crazy number of Grand Prix races or beating so many people in online races.

Cups
Star Cup: Complete the Mushroom Cup and Flower Cup with trophy wins.
Leaf Cup: Complete the Shell Cup and Banana Cup with trophy wins.
Special Cup: Complete the Star Cup with a trophy win.
Lighting Cup: Complete the Leaf Cup with a trophy win.
Characters
King Boo: Beat the Star Cup at 50cc.
Diddy Kong: Beat all the cups at 50cc.
Baby Daisy: Get at least a 1 Star ranking in all the Wii Grand Prix Cups at 50cc.
Dry Bones: Beat the Leaf Cup at 100cc.
Mii Character: Beat the Special Cup at 100cc.
Bowser Jr.: Get at least a 1 Star ranking in all the Retro Grand Prix Cups at 100cc.
Daisy: Beat the Special Cup at 150cc.
Dry Bowser: Get at least a 1 Star ranking in all the Wii Grand Prix Cups at 150cc.
Birdo: Either play on 16 different courses in Time Trial mode or finish ahead of a total of 250 players online.
Rosalina: You will need to have a Super Mario Galaxy save file on your Wii and the game will award you with this character after playing around for a bit. It seems you can unlock it too by finishing all Retro Tournaments, Mirror Mode, with at least one star.
Toadette: Play on all 32 courses in Time Trial mode.
Funky Kong: In Time Trial mode, unlock 4 Expert Staff Ghost Datas.
Baby Luigi:In Time Trial mode, unlock 8 Expert Staff Ghost Datas.
Mii Character (Mario Costume) : In Time Trail mode, unlock all 32 Expert Staff Ghost Data.
Karts
Piranha Prowler: (Large) Beat the Special Cup at 50cc.
Super Blooper: (Medium) Beat the Leaf Cup at 50cc.
Cheep Charger: (Small) Get at least a 1 Star ranking in all the Retro Grand Prix Cups at 50cc.
Daytripper: (Medium) Beat the Leaf Cup at 150cc.
Honeycoupe: (Large) Beat the Lightning Cup at 150cc.
Blue Falcon: (Small) Beat the Lightning Cup on Mirror Mode.
Tiny Titan: (Small) In Time Trial mode, unlock 1 Expert Staff Ghost Data.
Jetsetter: (Large) Get at least a 1 Star ranking in all the Retro Grand Prix Cups at 150cc.
Sprinter (Medium) In time Trials mode, unlock 24 Expert Staff Ghost Data.
Bikes
Zip Zip: (Medium) Beat the Lightning Cup at 100cc.
Shooting Star: (Large) Beat the Star Cup at 100cc.
Sneakster: (Medium) Get at least a 1 Star ranking in all the Wii Grand Prix Cups at 100cc.
Quacker: (Small) Beat the Star Cup at 150cc.
Dolphin Dasher: (Medium) Beat the Star Cup in Mirror Mode.
Phantom: (Large) Beat the Special Cup in Mirror Mode.
Bubble Bike: (Small) Beat the Leaf Cup in Mirror Mode.
Maglkruiser: (Small) Play in Time Trial mode on 8 courses.
Spear: (Large) In Time Trial mode, unlock 12 Expert Ghost Data.
Mirror Mode
Earn trophies in all 150cc cups.
