Critical Hits
While most of the creatures in the game have "weak-spots," the critical hits that go towards your end score are only for zombies and crimson heads. The zombie's critical hit point is the head, but not just anywhere on the head. Their weak-spot is somewhat hard to find at first. The best way to find it nearly every time is to aim for the top of their forehead. Try to hold your on-screen cursor almost just above their head. You should see the target focus and blink red when you have the right spot.
It's almost best to play sitting up with your aiming arm resting completely on your leg. The only thing that should be slightly suspended in the air is your hand and part of your wrist. Resting your arm on your leg should stabilize your aiming quite a bit. Now find the sweet (critical hit) spot on-screen and let the zombies simply walk into your cursor at head level and you can just pop one after the other. Know that if you are having to shoot a zombie in the head a couple times, you are not hitting the weak-spot. Any zombie and crimson head will go down with one critical hit.
