Evolution of Humans
This link should explain a bit of it, and of course you can go deeper into the meaning. 46 and 2 is referring to the number of chromosomes evolved humans would have based on a few theories, and thus the song is about looking forward to the next evolutionary phase. -- Colddweller

edit: Jambi is definitely up there; it's a thousand times better when they open their concerts with it too. Lateralus is my personal fav. I suppose that's because it's a heck of a lot more fun to play than Jambi is. Now if I had a talk box that might be different...
